Suicidality and Antidepressant Drugs
Antidepressants increased the risk compared to placebo of suicidal thinking and behavior (suicidality) in children, adolescents, and young adults in short-term studies of Major Depressive Disorder (MDD) and other psychiatric disorders. Anyone considering the use of venlafaxine tablets or any other antidepressant in a child, adolescent, or young adult must balance this risk with the clinical need. Short-term studies did not show an increase in the risk of suicidality with antidepressants compared to placebo in adults beyond age 24; there was a reduction in risk with antidepressants compared to placebo in adults aged 65 and older. Depression and certain other psychiatric disorders are themselves associated with increases in the risk of suicide. Patients of all ages who are started on antidepressant therapy should be monitored appropriately and observed closely for clinical worsening, suicidality, or unusual changes in behavior. Families and caregivers should be advised of the need for close observation and communication with the prescriber. Venlafaxine tablets are not approved for use in pediatric patients (see WARNINGS: Clinical Worsening and Suicide Risk, PRECAUTIONS: Information for Patients, and PRECAUTIONS: Pediatric Use).

Overview
Venlafaxine tablets are an antidepressant medicine used to treat major depressive disorder in adults. The label says its effectiveness was shown in short-term controlled trials and in a longer-term trial for helping prevent relapse in patients who responded to treatment.
The label warns that antidepressants can increase the risk of suicidal thoughts or behavior in children, adolescents, and young adults, and that people of all ages should be watched closely for worsening depression, unusual behavior changes, or suicidality, especially when starting treatment or changing the dose. Venlafaxine tablets are not approved for pediatric use.
Venlafaxine should not be used with certain medicines called MAOIs, and it can cause serotonin syndrome, a potentially life-threatening reaction, especially when combined with other serotonergic drugs. The label also warns about sustained increases in blood pressure, possible angle-closure glaucoma in people with narrow angles, sexual dysfunction, and symptoms that may happen if the medicine is stopped suddenly.

- Take venlafaxine exactly as prescribed on the label.
- Do not start or stop venlafaxine without checking with the prescriber, because stopping suddenly can cause withdrawal-like symptoms.

- Blood pressure should be monitored during treatment because venlafaxine can cause sustained increases in blood pressure.
- Report eye pain or vision changes promptly, since antidepressants like venlafaxine may trigger angle-closure glaucoma in people with narrow angles.
- Tell the prescriber about any sexual side effects, because sexual dysfunction can occur with SNRIs like venlafaxine.
- If a dose is being reduced or the medicine is being stopped, the label recommends gradual tapering when possible.
- Keep follow-up appointments so the prescriber can review how well the medicine is working and watch for side effects.
