Suicidality and Antidepressant Drugs:
Antidepressants increased the risk compared to placebo of suicidal thinking and behavior (suicidality) in children, adolescents, and young adults in short-term studies of major depressive disorder (MDD) and other psychiatric disorders. Anyone considering the use of amitriptyline hydrochloride tablets or any other antidepressant in a child, adolescent, or young adult must balance this risk with the clinical need. Short-term studies did not show an increase in the risk of suicidality with antidepressants compared to placebo in adults beyond age 24; there was a reduction in risk with antidepressants compared to placebo in adults aged 65 and older. Depression and certain other psychiatric disorders are themselves associated with increases in the risk of suicide. Patients of all ages who are started on antidepressant therapy should be monitored appropriately and observed closely for clinical worsening, suicidality, or unusual changes in behavior. Families and caregivers should be advised of the need for close observation and communication with the prescriber. Amitriptyline hydrochloride is not approved for use in pediatric patients. Overview
Amitriptyline hydrochloride tablets are a prescription antidepressant used for the relief of symptoms of depression. The label notes that endogenous depression is more likely to be relieved than other depressive states.
The medicine has sedative effects. It is not a monoamine oxidase inhibitor and does not act primarily by stimulating the central nervous system.

Storage Information
Protect the patient's airway, establish an intravenous line and initiate gastric decontamination. Store at 20º to 25ºC (68º to 77ºF); excursions permitted to 15° to 30°C (59° to 86°F) [See USP Controlled Room Temperature].

- Tell the prescriber right away about agitation, irritability, insomnia, hostility, impulsivity, hypomania, or mania.
- Do not take amitriptyline with monoamine oxidase inhibitors; the label says the combination is contraindicated.
- Report symptoms that could suggest serotonin syndrome, such as agitation, hallucinations, fever, tremor, rigidity, or diarrhea.
- Use caution with alcohol and other CNS depressants, since the medicine may increase their effects.
- Tell the prescriber about a history of seizures, urinary retention, or angle-closure glaucoma.
- Use caution if you have cardiovascular disorders, and report palpitations or other concerning symptoms.
- This medicine may affect alertness; be careful with driving or operating machinery until you know how it affects you.
